Definitely a trip to the vet is in order. This is not normal, and not healthy.

Do, as was suggested, clean and deodorize that spot. Stick a piece of furniture there if you have to so he cannot get to that spot.

Are there any other changes - how long have you had his "sister"? Anything that might be stressing him out?
